The present invention relates to two peptide nucleic acid (PNA) probe systems having a parallel binding structure for real-time amplification of nucleic acid, which can be nested-hybridized with a target nucleotide, a composition comprising the probe system, and a method for detection using same. By using the composition comprising the PNA probe system, according to the present invention, the existence of a nucleotide to be detected can be confirmed, the amount of the nucleotide can be decided at a fixed quantity, single nucleotide polymorphism (SNP), which is difficult to detect with existing technique, can be detected with high sensitivity, and simultaneous multiplexed detection of nucleotides of at least two types existing in a sample can be possible.
