The present invention relates to human interleukin 12 (IL12) gene expression regulating sequences, including the IL-12p35 subunit gene promoter sequence, and the first intron sequence of the human IL-12p40 subunit gene, preferably operably linked to the first exon of the human IL-12p40 subunit gene and further upstream to the human IL-12p40 subunit gene promoter sequence. Further, the invention relates more particularly to DNA constructs containing such a sequence and a reporter gene, to vectors and host cells containing such a DNA construct, to a method of screening for compounds which affect the production of IL-12 in such cells, in particular human cells, to non-proteinaceous compounds obtainable by such a method, and to transgenic non-human mammalian animals which express a reporter gene under the control of one of said IL-12 gene expression regulating sequences.
