本發明係關於一種目標蛋白質之生產方法及表現目標蛋白質之懸浮性之哺乳動物細胞，該目標蛋白質之生產方法係將於包含編碼目標蛋白質之DNA及選擇標記基因之基因片段之兩端含有轉位子序列的蛋白質表現載體導入至懸浮性之哺乳動物細胞中，並將插入在一對轉位子序列間的該基因片段組入於該哺乳動物細胞之染色體中而獲得表現該目標蛋白質之哺乳動物細胞，且對該哺乳動物細胞進行懸浮培養，從而生產該目標蛋白質。

本发明系关于一种目标蛋白质之生产方法及表现目标蛋白质之悬浮性之哺乳动物细胞，该目标蛋白质之生产方法系将于包含编码目标蛋白质之DNA及选择标记基因之基因片段之两端含有转位子串行的蛋白质表现载体导入至悬浮性之哺乳动物细胞中，并将插入在一对转位子串行间的该基因片段组入于该哺乳动物细胞之染色体中而获得表现该目标蛋白质之哺乳动物细胞，且对该哺乳动物细胞进行悬浮培养，从而生产该目标蛋白质。

Disclosed is a process for producing a protein of interest, which comprises: introducing, into a floating mammalian cell, a protein expression vector that comprises a gene fragment comprising DNA encoding the protein and a selection marker gene and transposon sequences attached to both termini of the gene fragment to integrate the gene fragment inserted between the pair of transposon sequences into the chromosome of the mammalian cell, thereby producing a mammalian cell capable of expressing the protein; and carrying out the floating culture of the mammalian cell to produce the protein. Also disclosed is a floating mammalian cell capable of expressing a protein of interest.
