The present invention provides compositions, apparatus and methods for detecting one or more nucleic acid target present in the sample. The method of the present invention reversibly binding to the target nucleic acid that approaches close to each other and comprising the step of using two or more ligation probes possessing a complementary reactive moiety ligation. When such a probe is to be bonded to the target in the proper orientation, they may perform spontaneous chemical ligation reaction which generates a ligation product, and this is the ligation product is detected directly, or amplified to generate the amplicons is detected thereafter. The present invention also provides a method to prevent the degradation by stabilizing the RNA samples not have a significant effect on the analysis results.
