PROBLEM TO BE SOLVED: To provide new methods and means for isolation and purification of a nucleic acid on the surface. SOLUTION: A method using the surface, e.g., a porous film, immobilizing with a simple method a nucleic acid from a sample including the nucleic acid, also releasing with a simple process and thereby specifically enabling practice of a perfectly automated method, is provided. And further, in the subsequent reaction process, the nucleic acid is released from the phase without complexity again and the nucleic acid is used, e.g., for restriction digestion, RT, PCR, or RT-PCR or in another analytical or enzymatic reaction.
