The invention provides a tumor vaccine and a preparation method thereof. The tumor vaccine contains dendritic cells blocking CD11b molecular signaling pathways. The invention further provides the method for preparing the tumor vaccine. The method comprises the steps that (1) mononuclear cells are obtained from peripheral blood of mammals and induced into mature dendritic cells, or bone marrow hematopoietic precursor cells are obtained from bone marrows of the mammals and induced into mature dendritic cells; (2) the CD11b molecular signaling pathways of the mature dendritic cells are blocked; and (3) the dendritic cells blocking the CD11b molecular signaling pathways are collected. The provided tumor vaccine has the good anti-tumor effect, meanwhile, is wide in anti-tumor spectrum, and hasgood using safety.
