The method comprises the following steps: cloning aCADl-A gene to obtain CADl-A-l plasmids; constructing an RNAi expression vector of the CADl-A gene; performing genetic transformation on embryonic callus of transgenic Bt insect-resistant cotton by adopting an agrobacterium tumefaciens mediation method, wherein the CADl-A gene in the RNAi expression vector is driven by an alpha-globulin promoter, and the alpha-globulin promoter is modified with an Enh I enhancer. The method can inhibit methylation of the Bt gene promoter and the CADl-A gene seed specific promoter, so that inhibition on the Bt gene transcription level in a compound transgeniccrop can be remarkably reduced, reduction of the CADl-A gene transcription level can be promoted, toxin on embryonic callus can be reduced, the programmed death of transformed cells can be inhibited,and the transformation frequency can be improved.
