The invention relates to a method for evaluating risk exposure on an animal body by tobacco-specific nitrosamines. The method comprises the following steps: 1) detecting content of TSNAs and metabolite thereof; computing an AUC value, computing an alpha-hydroxylation coefficient according to the AUC value, and detecting content of the O6-mG and O2-POB-dT; and acquiring the yield of the representative DNA adduct according to the sum of the content values of the O6-mG and O2-POB-dT; 2) evaluating the alpha-hydroxylation metabolic degree according to the alpha-hydroxylation coefficient and the representative DNA adduct yield, and predicting the exposure risk on the animal body by the TSNAs. Through the method disclosed by the invention, the alpha-hydroxylation reaction degree in different individuals after the TSNAs is exposed can be evaluated according to the alpha-hydroxylation coefficient and the yield of the representative DNA adduct, thereby providing quantitative index for the potential exposure risk of the TSNAs.
