The invention, which relates to the field of biotechnology, particularly discloses a biosensor for detecting the Norovirus and a preparation and use method thereof. The biosensor comprises a working electrode being cadmium-sulfide-quantum-dot-coated indium tin oxide; a specific antibody and/or a specific probe is coupled to the cadmium sulfide quantum dot surface; and the specific antibody is onedesigned specially for the nucleocapsid protein VP1 of the Norovirus or the specific probe is one designed for the Norovirus. According to the biosensor formed simply, no complicated technology and equipment are involved and the requirements on technologies and equipment are low; and the detection sensitivity is high. The biosensor can be applied widely.
