The invention discloses a canine distemper virus (CDV) antibody fluorescence test strip and a preparation method and an application thereof. The test strip comprises a sample absorption pad, a conjugate release pad a, a conjugate release pad b, a nitrocellulose membrane, a water absorption pad and a bottom board. An anti-chicken anti-dog antibody-biotin conjugate is sprayed on the conjugate release pad a; streptavidin-fluorescent microsphere markers are sprayed on the conjugate release pad b; and an inspection line and a quality control line are formed on the nitrocellulose membrane. The inspection line is coated with canine distemper virus N expression protein; and the quality control line is coated with an anti-goat anti-chicken antibody. The test strip has the advantages of being high in sensitivity, high in specificity, simple to operate and economical and practical and the like, and can realize rapid and quantitative on-site test of the CDV antibody.
