The invention relates to the field of drilling of petroleum industry, in particular to a nano silica-grafted copolymer with abilities to tolerate temperature and calcium and suitable for reducing filtrate loss, a preparation method and application thereof, a drilling fluid and application of the drilling fluid. The nano silica-grafted copolymer comprise one or more of structural units shown as formula (1), one or more of structural units shown as formula (2), one or more of structural units shown as formula (3), and one or more of structural units shown as formula (4). The nano silica-graftedcopolymer provided herein can successfully provide good resistance to high temperature and high calcium when used in the drilling fluid as a filtrate loss reducer through the cooperation of the structural units shown as the formula (1), the structural units shown as the formula (2), the structural units shown as the formula (3) and the structural units shown as the formula (4).
