The invention provides a single-hole vacuum oiling device of an oil-immersed transformer, and an oiling method based on the device. The single-hole vacuum oiling device comprises a vacuum pump group,a water-gas separator, an oil storage tank, a vacuum oil filter group, an oil storage tank valve and a three-way pipe, the vacuum pump group comprises at least one vacuum pump, each vacuum pump is separately connected with a vacuum pump valve, the vacuum pump group is sequentially connected with the water-gas separator, the three-way pipe and a transformer check valve, a transformer oil conservator is connected with an oil conservator valve, the oil conservator valve is connected with the three-way pipe, and the oil storage tank is sequentially connected with the oil storage tank valve, the vacuum oil filter group and a transformer oil injection and discharge valve. The single-hole vacuum oiling device provided by the invention is simple in oil injection and can avoid the oxidation of thetransformer oil.
