According to the preparation method, chicken serum is used for replacing bovine serum, and harmful stimulus on chicken cells by heterologousserum can be avoided; the inducing culture medium and the enhanced culture medium are used for carrying out inducing culture on the single karyocyte in two stages, lactoalbumin hydrolysate and chickembryo allantoic fluid are added into the enhanced culture medium, so that the requirements of later stage cell proliferation on nutrition are met, and meanwhile, the proliferation and the differentiation of cells are accelerated.
