The invention relates to a seamless steel pipe for a non-excavation integral drilling rod and a preparation method of the seamless steel pipe. The seamless steel pipe comprises the following ingredients by mass percentage: 0.27-0.32% of C, 0.15-0.35% of Si, 1.60-2.00% of Mn, 0.8-1.8% of Cr, 0.20-0.26% of V and the balance of Fe and unavoidable impurities. In a thermal refining process, a quenchingtemperature is controlled at 810-830 DEG C, and a tempering temperature is controlled at 570-650 DEG C. On the premise of ensuring mechanical properties of the drilling rod meet standards, the addition of Mo element is avoided, and the production cost is lowered by regulation of contents of Mn and Cr in the seamless steel pipe and through a subsequent quenching and tempering technology. The prepared steel pipe fully meets various properties of an S135 steel grade specified in API (American Petroleum Institute) 5DP, and at the same time, the steel pipe has good price advantages, economic benefits and application prospects.
