The invention discloses a separation and purification method of malvidin-3-O-galactoside and application thereof in preparing an alpha-glucosidase inhibitor. The separation and purification method comprises the following steps: by taking blueberries with complex anthocyanin composition as a raw material, performing alcohol extraction and concentration, macroporous resin adsorption, preparative liquid chromatography purification and high-speed counter-current chromatography separation, thereby obtaining the high-purity malvidin-3-O-galactoside monomer by technically combining preparative liquidchromatography with high-speed counter-current chromatography, optimizing process parameters and carrying out separation, wherein the purity is as high as 99%. An activity test shows that the malvidin-3-O-galactoside monomer can significantly inhibit the activity of alpha-glucosidase and can be used for preparing the alpha-glucosidase inhibitor.
