The method comprises the steps of S1 preparing a culture solution for storing male fish semen, obtaining the male fish semen and using the culture solution for constant volume according to the ratio being 1:(30-40) to obtain a mixed solution, and dividing the mixed solution into multiple parts; S2 preparing an oxygen enrichment bottle containing sufficient oxygen, and subpackaging and refrigerating the mixed solution in step 1 into the oxygen enrichment bottle; S3 preparing a lubricant, and adding 10 ml of lubricant into each insemination unit so as to perform cleaning and lubricating; S4 after cleaning and lubricating are performed, adding one part of mixed solution in S2 into each insemination unit, performing sufficient stirring and mixing, then adding 450-550 ml of heavy-oxygen-enriched water for stirring, and completing insemination. The quantified operation of artificial insemination is achieved, the mixed solution containing male fish semen has the long shelf life, and meanwhile the utilization efficiency of wild parent male fish in the artificialendangered fish breeding process is improved by 300-400 times.
