The invention relates to a preparation and quality detection method of traditional Chinese medicine extract, in particular to a preparation and quality detection method of amomum para-tsaoko total flavonoids extract. The method comprises the following steps: crushing and alcohol extracting dry fruits of amomum para-tsaoko, performing reflux extraction with methanol or ethanol, concentrating, adding water, mixing, suspending, eluting by using a macroporous adsorption resin column chromatograph (ethanol-water system), concentrating, vacuum drying, and obtaining the amomum para-tsaoko total flavonoids extract. The preparation method of the amomum para-tsaoko total flavonoids extract is reasonable, stable, short in production period, and suitable for industrialized production. The invention adopts an ultraviolet-visible light spectrophotometry, a high-performance liquid-phase chromatographic one-measuring multi-evaluation method and an area normalization method to determine the content of amomum para-tsaoko total flavonoids in the amomum para-tsaoko total flavonoids extract. The method is accurate, stable, and simple in operation. The amomum para-tsaoko total flavonoids extract prepared by the invention has pharmacologic actions such as inflammation resistance, bacterial resistance and the like.
