The invention discloses a preparation method of a 2-methoxyl-5-geranyl p-benzoquinone derivative. The method is implemented by the following steps: 1, preparing a middle product 1,2,4-trimethoxy-3-trimethylsilyl-benzene; 2, preparing a middle product 1,2,4-trimethoxy-6-geranyl-benzene; 3, preparing a target. The invention further discloses a preparation method of Scabellone C. According to the preparation methods, 1,2,4-trimethoxy benzene is used as a raw material, so that the 2-methoxyl-5-geranyl-p-benzoquinone derivative (a coupled product) can be successfully obtained by one step through ammonium ceric nitrate oxidization under a common organic solvent condition, and the Scabellone C is synthesized under a condition of selecting different organic solvents; therefore, a breakthrough for zero yield in the aspect of preparation of the Scabellone C in the prior art is realized; meanwhile, by the adoption of the preparation method disclosed by the invention, the yield of the Scabellone C is extremely high.
