The invention relates to a triptolide derivative, and a preparation method and use thereof. The triptolide derivative is a compound having the structure as shown in a formula (I) and a formula (II), or a medically acceptable salt of the compound having the structure as shown in the formula (I) and the formula (II), or an optical isomer of the compound having the structure as shown in the formula (I) and the formula (II), or an optical isomer of the medically acceptable salt of the compound having the structure as shown in the formula (I) and the formula (II). The triptolide derivative provided by the invention can efficiently inhibit the function of the spongiocyte BV2 to secrete TNF-alpha at solubility of pM (10-12mol/L), and meanwhile, the compound can inhibit the toxicity of A beta-42 to the primary cortical neurons.
