The invention discloses a detection method and particularly relates to a method of detecting B vitamins in food. The method comprises the steps of (1) pretreating a food sample to obtain a loading liquid; (2) analyzing by HPLC-MS/MS (high performance liquid chromatography-mass spectrometry and mass spectrometry), to be specific, measuring both a B vitamin standard solution and the loading liquid of step (1) by means of HPLC-MS/MS, comparing measurement results of the loading liquid with measurement results of the B vitamin standard solution, and qualifying or quantifying B vitamins in the food sample. The method provides high separation degree of B vitamins and good separation effect; the method has high sensitivity and accuracy and low detection limit for the detection of B vitamins, and is suitable for detecting more than 8 sorts of B vitamins.
