The invention discloses a microRNA detection method based on in-situ generation of Cu-nanoclusters from thymine formed through extension of double polymerases, belonging to the field of optical biosensing technologies. Based on the co-catalysis of polymerase Klenow fragments and terminal deoxynucleotidyl transferase, when microRNA exists, deoxythymidine triphosphate is arranged and extended to form a long poly-thymine base sequence, and red fluorescence Cu-nanoclusters are generated through in-situ reaction by taking the poly-thymine base sequence as a template, wherein the fluorescence intensity of the Cu-nanoclusters is in direct proportion to the logarithm of the concentration of microRNA, and thus the low-background and high-sensitivity detection for microRNA is realized.
