The invention discloses a lipidosome with entrapment nucleic acid drugs, an anaerobic response capability and a radiotherapy sensitivity enhancing function. The lipidosome comprises a lipidosome body and nucleic acid drugs. The lipidosome is prepared from, by mass, 20-80% of lipid molecules with the radiotherapy sensitivity enhancing function, 5-50% of cholesterol, 3-50% of polyethylene glycol phospholipid and 1-30% of DOPC lipid molecules. The N/P ratio of the lipidosome to nucleic acid in the nucleic acid drugs is 1:(1-20). The obtained lipidosome overcomes the defects that the positive ion lipidosome toxicity is high, the blood stability is poor and the transfection efficiency is low, the effect of the nucleic acid drugs is effectively improved, meanwhile the lipidosome has the high endosome escape capability under the oxygen-deficient environment, so that the obvious advantage of transfection of gene-based drugs is achieved. The lipidosome is applicable to entrapment of various nucleic acid drugs, and the applicable dosage form range is wide.
