The invention relates to a preparation method for a polymorphic bio-polypeptide organic fertilizer. The preparation method comprises the following steps: with high-protein content peanut straw, domestic fungus culture medium waste residue, wheat bran and soybean meal as raw materials, carrying out water extraction so as to extract protein in the raw materials; and successively adding an alkali protease preparation, a neutral protease preparation and an acidic protease preparation and carrying out three-level decomposition so as to prepare the polymorphic bio-polypeptide organic fertilizer. The preparation method does no need complex and energy-consuming after-treatment procedures like separation and purification, so production cost of the polypeptide organic fertilizer is substantially reduced; and since the polypeptide organic fertilizer contains polymorphic polypeptides, the fertilizer has improved fertilizer efficiency, and field test results show that the fertilizer has good yield increasing effect.
