The invention belongs to the field of biological products and provides a coding gene of salmonella typhimurium outer membrane protein L, a transfer vector pUSG11/P28-OmpL with the coding gene, a recombinant swinepox virus rSPV-OmpL and application and a preparation method of the recombinant swinepox virus rSPV-OmpL. The sequence of the coding gene is shown as SEQ ID NO.1, and the recombinant swinepox virus is obtained by insertion of the salmonella typhimurium outer membrane protein L into swinepox virus VR-363. A recombinant swinepox virus vaccine rSPV-OmpL is capable of greatly expressing target protein OmpL in immune animal bodies to induce in-vivo generation of high-potency antibodies aiming at OmpL in the immune animal bodies, the immune animals can be well protected, and the immune protective rate is up to 80% as for mice. In addition, the vaccine integrates advantages of excellent immune effects of attenuated vaccines and high safety of inactivated vaccines, thereby being an ideal novel vaccine.
