The method specifically comprises the following steps: adding 60-100mg of tobacco in every 1ml of tobacco seed grease, and mixing uniformly; coating a glass slide with a heated solid culture medium by using a glass slide sheet; sucking 10mul of the tobacco seed grease containing the tobacco pollen by using a liquid transfer gun, putting the tobacco seed grease on a cooled solid culture medium, and coating uniformly by using a glass rod; and putting the glass slide into a culture dish in which two layers of moistened filter paper are added, then putting the culture dish into an incubator to perform dark culture at 25 DEG C, observing the germination condition of the pollen under a microscope after 4 hours, and counting the activity of the pollen. The pollen in the method disclosed by the invention has good dispersity in the tobacco seed grease, is not superimposed when coated on the solid culture medium, and is convenient to count and count the activity of the pollen; and the tobacco seed grease can promote the germination of the pollen, shortens the time for detecting the activity of the pollen, and improves the efficiency of detecting the activity of the pollen.
