The invention discloses a method for preparing a pristinamycin IIA single component. The method comprises the following steps: a, eluting pristinamycin IIA crude product by adopting a binary liquid phase separation system and anti-phase filling material chromatography through a brine solution containing organic solvent, and collecting eluant of which the pristinamycin IIA purity is greater than 98.5%; b, conducting concentration, filtration and drying on the eluant obtained in the step a, so as to obtain the pristinamycin IIA single component. The method adopts the technical means of industrial chromatographic separation so as to obtain the pristinamycin IIA single component, the process is concise, the quality is controllable, and the industrialized production is facilitated.
