The invention discloses a constructing method of a stable self-luminous recombinant vector and an application of the recombinant vector. The recombinant vector is obtained by inserting a chloroplast rps7 gene upstream homologous arm, a fatty acid reductase encoding gene, a transferase encoding gene, a luciferase alpha-subunit encoding gene, a luciferase beta-subunit encoding gene, a synthetase encoding gene, a flavin reductase encoding gene and a chloroplast ndhB gene downstream homologous arm into a chloroplast expression vector. Two sequences derived from the poplar (populus davidiana) chloroplast DNA on the recombinant vector are taken as the homologous recombinant fragments and the luminous gene is inserted into a spacer region between the poplar chloroplast genome rps7 gene and ndhB gene in a fixed point, so that the luminous gene is effectively expressed in the poplar chloroplast genome, the self-luminous transgenosis poplar is obtained and the application prospect is wonderful.
