The invention provides a method for positioning and transplanting tobacco seedlings. The method comprises a positioning step based on a positioner, a ridge forming step, a film covering step and a tobacco seedling transplanting step which are sequentially performed, wherein the positioner is a round wood rod or a similar object and has length of 35-40cm and diameter of 8-10cm, one end of the positioner is a sharp end which is similar to a cone and the height of the cone is 8-10cm, and the positioner is inserted into soil before ridge forming; during ridge forming, the height of a ridge body is 25-30cm, and the positioner is 3-5cm higher than the ridge body; in the film covering step, a hole formed by pulling out the positioner after ridge forming is covered with a film, and the film is kept tensioned; in the tobacco seedling transplanting step, the film at the upper side of the hole is torn to form 3-4cm holes into which the tobacco seedlings are put. According to the method, more suitable humiture and nutritive water conditions are provided for the tobacco seedlings, all transplanting operation procedures are effectively combined, the problems that traditional transplanting is labor-consuming and time-consuming, and early rooting and quick sprouting after transplanting are not ideal, and the like are solved, and the cured tobacco transplanting effect is improved.
