本发明提供一种S-腺苷甲硫氨酸脱羧酶的药物口袋,其特征在于:以S-腺苷甲硫氨酸脱羧酶(AdoMetDC)的晶体结构为基础,将PDB ID为3DZ5的AdoMetDC晶体结构中的第68位丙酮酰基用PDB ID 1JL0的AdoMetDC晶体结构中的第68位色氨酸残基替代,以替代后的AdoMetDC晶体结构为基础,通过Pocket蛋白质药物口袋分析软件,确定在alpha亚基和beta亚基两条链之间形成的药物口袋为S-腺苷甲硫氨酸脱羧酶的药物口袋,并将该药物口袋应用于AdoMetDC抑制剂的筛选、设计和改造。

The invention provides a drug pocket of AdoMetDC (S-adenosylmethionine decarboxylase). The drug pocket is characterized in that on the basis of an AdoMetDC crystal structure, the 68-poistion pyruvoyl in the AdoMetDC crystal structure whose PDB (protein data bank) ID is 3DZ5 is substituted by the 68-poistion tryptophan residue in the AdoMetDC crystal structure whose PDB ID is 1JL0; and on the basis of the AdoMetDC crystal structure after substitution, the condition that a drug pocket formed between two strands of alpha subunit and beta subunit is the drug pocket of the AdoMetDC is determined by the pocket protein drug pocket analysis software, and the drug pocket is applied to screening, design and transformation of an AdoMetDC inhibitor.
