The invention relates to a kit for detecting the mutation of a gene Fibulin-5 of sika deer. The kit comprises a reagent for amplifying the gene Fibulin-5 and a reagent for detecting the mutation of the gene Fibulin-5. The reagent for amplifying the gene Fibulin-5 comprises PCR (Polymerase Chain Reaction) primers for amplifying the gene Fibulin-5 and corresponding PCR reagents, wherein the PCR primers for amplifying the gene Fibulin-5 are used for detecting whether the 1,030-th and 1,081-th nucleotides of the gene Fibulin-5 of the sika deer are subjected to C-to-T mutation or not. Sequences of the PCR primers comprise F778: 5'-ACACCGAAGGAGGCTACAC-3' and R1307: 5'-CTCCTCACAGCGGATGGG-3'. According to the kit, found by researches, when the 1,030-th and 1,081-th nucleotides of the gene Fibulin-5 of the sika deer are subjected to C-to-T mutation, amino acid (cysteine) coding the protein of the gene Fibulin-5 is not changed, and the situation is related to the cutis laxa of the rear neck of the sika deer. The primers, method and kit disclosed by the invention have the advantages of relatively high amplification stability, specificity, high sensitivity, high accuracy and the like.
