The invention discloses a conditioner for fermenting an organic fertilizer from sludge and manufacturing and using methods thereof. The conditioner comprises the following components in weight proportion: 30-35 parts of soybean cake powder, 65-70 parts of bran, 1-2 parts of sugar, 0.5-1 part of KH2PO4 and 1*10<7>-1*10<9> pieces of bacillus natto per gram of culture medium. In the invention, the powerful pathogenic bacterial restrain capacity of the bacillus natto is utilized to produce acid and regulate flora, and by means of sludge fermentation and the bacillus natto, the sludge is made harmless. The bacillus natto can also be used for generating various proteases (particularly alkali protease), diastatic enzyme, lipase and amylase, and degrading certain complex carbohydrates of vegetable matters, so that the conversion rate of the sludge is increased.
