The invention discloses a palladium-catalyzed asymmetric propyl alkylation reaction method in the technical field of chemical engineering. In solvent, under the catalysis of an in-situ catalyst formed by a chiral ligand and a palladium catalyst, substituted allyl acetic ester and in-situ enamine are enabled to be subjected to an alkylation reaction, wherein the in-situ enamine is formed by aldehyde or ketone and secondary amine in situ. In the invention, the in-situ enamine is used as a nucleophile in an asymmetric catalytic reaction, thereby instable enamine is prevented from being synthesized and the palladium-catalyzed asymmetric propyl alkylation reaction method is simple, convenient and safe for operation. In addition, the palladium-catalyzed asymmetric propyl alkylation reaction method has favorable reaction yield and reaction efficiency as well as better application effect.
