The invention discloses a method and a kit for detecting carbon dioxide bonding force. The detecting method comprises the following steps of: mixing a sample to be detected of carbon dioxide bonding force and detection solution, and performing reaction to obtain reaction solution, wherein the detection solution comprises Tris-HCl buffer solution with pH value of 4.8 to 5.2, sodium chloride, carbonic anhydrase and color development agent; and detecting the absorbance value of the reaction solution under the wavelength of 540 to 550 nanometers, and obtaining the carbon dioxide bonding force by calculating. In the detection process, the carbon dioxide in the sample to be detected generates bicarbonate radicals under the action of the carbonic anhydrase, so that the pH value of the reaction solution is raised, and the color development agent develops color; and the carbon dioxide bonding force is calculated by detecting the absorbance value of the reaction solution. Compared with the prior art, color development reaction is used, reductive coenzyme is not needed, and the detected carbon dioxide bonding force is accurate.
