The invention relates to a kit and a method for detecting polymorphism of an FMO3 gene of an egg-laying chicken, belonging to the biotechnology field. The kit comprises a forward primer which has a base sequence of 5'-CGGTGTTGAATGATGAC-3', a reverse primer which has a base sequence of 5'-GCATTTGTAGAGGGTAG-3', dNTPs, DNA polymerase and a 10*PCR buffer solution thereof and restriction endonuclease BsrI and a corresponding 10*digestion buffer solution thereof. The invention solves the problem that the allele is difficult to eliminate by using the traditional breeding method, accelerates the breeding process, lowers the breeding cost and has extremely wide application prospects in the selection and breeding work of hybrids of non-fishy smell egg-laying chickens.
