The invention provides a telechelic polyethylene glycol with variant end-group, namely a variant end-group telechelic PEO and a preparation method of the variant end-group telechelic PEO. First, the invention uses ethyl vinyl ether to protect hydroxyl at one end of diol HO-(CH2)n-OH,n=2-10 or HO-(CH2-CH2-O)n-H,n=1-5. Then the production obtained through the above process has reaction with metal potassium to prepare small molecule initiator which can successfully initiate the ring-opening polymerization of ethylene oxide to make the PEO whose two ends are respectively protective hydroxyl and active hydroxyl. The active hydroxyl is modified first, and then the protective group at the other end of the hydroxyl is removed in the acid environment to make the variant end-group telechelic PEO; or the functional groups at the two ends of the PEO can be modified selectively to make various combination of the variant end-group telechelic PEO.
